    Study of metals in leached soils of a municipal dumpsite in Tampico, Tamaulipas, Mexico: preliminary results

    The Zapote dumpsite measures 420000 m 2 and is 28 years old; an estimated 2.5 millions tons of waste have accumulated on the site (household waste, clinical waste, commercial waste). The thickness of the waste is 3 to 9 meters. Since operations began, no control regulations have existed on the residues received. The Zapote dumpsite is located within a salt-marsh between a system of channels and river lagoons of brackish water, located in a tropical sedimentary environment in the urban zone of Tampico, Tamaulipas, Mexico. Recently, the Zapote has been closed and work is presently underway in its rehabilitation since a geo-environmental perspective. The present investigation integrates information of preliminary results of metals (Pb, Ni, Cd, Cu, Mg, Fe and Al) contained in sediments that underlie the Zapote dumpsite. In laboratory research the metals of the sediment were correlated with the metals contained in samples of leachate from the Zapote dumpsite. The concentration of metals Pb, Ni, Cd, Cu, Mg, Fe and Al were analyzed in samples of sediments that underlie the body of the dumpsite in layers of 10 cm, reaching a depth of 1.5 m under the interface waste-soil. The results denote high concentrations of metals in layers that are in contact with waste that decreased until reaching 60 to 80 cm of depth. The proportions of the concentrations of metals studied in the soil are comparable with that leached, until layers of 60 to 80 cm of depth are reached, and are then lost in the deepest layers. The high plastic characteristics of clay layers have stood in the way of metallic contaminants in sub layers of the Zapote dumpsite.     Deregulation of the imprinted DLK1-DIO3 locus ncRNAs is associated with replicative senescence of human adipose-derived stem cells

    Background Human adult adipose-derived stem cells (hADSCs) have become the most promising cell source for regenerative medicine. However the prolonged ex vivo expansion periods required to obtain the necessary therapeutic dose promotes progressive senescence, with the concomitant reduction of their therapeutic potential. Aim and scope A better understanding of the determinants of hADSC senescence is needed to improve biosafety while preserving therapeutic efficiency. Here, we investigated the association between deregulation of the imprinted DLK1-DIO3 region and replicative senescence in hADSC cultures. Methods We compared hADSC cultures at short (P S ) and prolonged (P L ) passages, both in standard and low [O 2 ] (21 and 3%, respectively), in relation to replicative senescence. hADSCs were evaluated for expression alterations in the DLK1-DIO3 region on chromosome 14q32, and particularly in its main miRNA cluster. Results Comparison of hADSCs cultured at P L or P S surprisingly showed a quite significant fraction (69%) of upregulated miRNAs in P L cultures mapping to the imprinted 14q32 locus, the largest miRNA cluster described in the genome. In agreement, expression of the lncRNA MEG3 (Maternally Expressed 3; Meg3/Gtl2), cultured at 21 and 3% [O 2 ], was also significantly higher in P L than in P S passages. During hADSC replicative senescence the AcK16H4 activating mark was found to be significantly associated with the deregulation of the entire DLK1-DIO3 locus, with a secondary regulatory role for the methylation of DMR regions. Conclusion A direct relationship between DLK1-DIO3 deregulation and replicative senescence of hADSCs is reported, involving upregulation of a very significant fraction of its largest miRNA cluster (14q32.31), paralleled by the progressive overexpression of the lncRNA MEG3, which plays a central role in the regulation of Dlk1/Dio3 activation status in mice.This work was supported by grants to AB from the Spanish Ministry of Economy, Industry (SAF2015-70882-R; AEI/FEDER, UE), Comunidad Autónoma de Madrid (S2010/BMD-2420), Instituto Salud Carlos III (RETICS TerCel, RD12/0019/0018) and the European Commission (FP7-HEALTH- 2009/CARE-MI).     Oxidized low-density lipoprotein receptor in lymphocytes prevents atherosclerosis and predicts subclinical disease

    Background: Although the role of Th17 and regulatory T cells in the progression of atherosclerosis has been highlighted in recent years, their molecular mediators remain elusive. We aimed to evaluate the association between the CD69 receptor, a regulator of Th17/regulatory T cell immunity, and atherosclerosis development in animal models and in patients with subclinical disease. Methods: Low-density lipoprotein receptor-deficient chimeric mice expressing or not expressing CD69 on either myeloid or lymphoid cells were subjected to a high fat diet. In vitro functional assays with human T cells were performed to decipher the mechanism of the observed phenotypes. Expression of CD69 and NR4A nuclear receptors was evaluated by reverse transcription-polymerase chain reaction in 305 male participants of the PESA study (Progression of Early Subclinical Atherosclerosis) with extensive (n=128) or focal (n=55) subclinical atherosclerosis and without disease (n=122). Results: After a high fat diet, mice lacking CD69 on lymphoid cells developed large atheroma plaque along with an increased Th17/regulatory T cell ratio in blood. Oxidized low-density lipoprotein was shown to bind specifically and functionally to CD69 on human T lymphocytes, inhibiting the development of Th17 cells through the activation of NR4A nuclear receptors. Participants of the PESA study with evidence of subclinical atherosclerosis displayed a significant CD69 and NR4A1 mRNA downregulation in peripheral blood leukocytes compared with participants without disease. The expression of CD69 remained associated with the risk of subclinical atherosclerosis in an adjusted multivariable logistic regression model (odds ratio, 0.62; 95% CI, 0.40-0.94; P=0.006) after adjustment for traditional risk factors, the expression of NR4A1, the level of oxidized low-density lipoprotein, and the counts of different leucocyte subsets. Conclusions: CD69 depletion from the lymphoid compartment promotes a Th17/regulatory T cell imbalance and exacerbates the development of atherosclerosis. CD69 binding to oxidized low-density lipoprotein on T cells induces the expression of anti-inflammatory transcription factors. Data from a cohort of the PESA study with subclinical atherosclerosis indicate that CD69 expression in PBLs inversely correlates with the presence of disease.     Science with Neutrino Telescopes in Spain

    [EN] The primary scientific goal of neutrino telescopes is the detection and study of cosmic neutrino signals. However, the range of physics topics that these instruments can tackle is exceedingly wide and diverse. Neutrinos coming from outside the Earth, in association with othermessengers, can contribute to clarify the question of the mechanisms that power the astrophysical accelerators which are known to exist from the observation of high-energy cosmic and gamma rays. Cosmic neutrinos can also be used to bring relevant information about the nature of dark matter, to study the intrinsic properties of neutrinos and to look for physics beyond the Standard Model. Likewise, atmospheric neutrinos can be used to study an ample variety of particle physics issues, such as neutrino oscillation phenomena, the determination of the neutrino mass ordering, non-standard neutrino interactions, neutrino decays and a diversity of other physics topics.     Nitrogen-carbon graphite-like semiconductor synthesized from uric acid

    A new carbon-nitrogen organic semiconductor has been synthesized by pyrolysis of uric acid. This layered carbon-nitrogen material contains imidazole-, pyridine (naphthyridine)- and graphitic-like nitrogen, as evinced by infrared and X-ray photoelectron spectroscopies. Quantum chemistry calculations support that it would consist of a 2D polymeric material held together by hydrogen bonds. Layers are stacked with an interplanar distance between 3.30 and 3.36 Å, as in graphite and coke. Terahertz spectroscopy shows a behavior similar to that of amorphous carbons, such as coke, with non-interacting layers. This material features substantial differences from polymeric carbon nitride, with some characteristics closer to those of nitrogen-doped graphene, in spite of its higher nitrogen content. The direct optical band gap, dependent on the polycondensation temperature, ranges from 2.10 to 2.32 eV. Although in general the degree of crystallinity is low, in the material synthesized at 600 °C some spots with a certain degree of crystallinity can be found

    Automatic imagine analyzer to assess retinal vessel caliber (Altair) tool validation for the analysis of retinal vessels

    Objective: To assess the reliability and validity of the tool Automatic image analyzer to assess retinal vessel caliber (ALTAIR) to analyze the vascularization of the retina and cardiovascular risk prediction. Design and method: Cross-sectional of tools validation study. We included 250 subjects in total, aged 62 +/- 9 years, 51% males. We have made a validation of reliability analyzing the intraclass correlation (ICC) intra observer, inter observer and inter device (compared with AVindex calculator) to the thickness, area and length of arteries and veins of the retina in 3 concentric circles from the disc in 120 retinographies. The concurrent validity was performed with 250 subjects and 497 retinographies, analyzing the relationship with age, blood pressure, parameters of vascular structure and function, renal function and cardiovascular risk estimated with scales. Results: In the sample, the 32% are obese, 68% hypertensive and 17% diabetic. The interobserver ICC for thickness, area and length of veins and arteries ranged from 0.809 to length of arteries to 0.916 to veins area. The intra observer ICC for intra thickness, area and length of veins and arteries ranged from 0.640 for the length of the veins and 0.906 for the area of the arteries and the inter device ICC was for arteriovenous ratio (AVR) 0.887, thickness of arteries 0.590 and veins thickness 0.677. We found a moderate correlation of the age (r between 0.30 and 0.50, p < 0.001) with the retinal vascular parameters analyzed. In multiple linear regression analysis after adjusting for age and sex, the association of AVR and arterial thickness with the diastolic blood pressure and albumin creatinine ratio and arterial area and length with systolic arterial blood pressure and carotid intima-media thickness remains. Also the thickness area and length of the vessels show an association with cardiovascular risk estimated SCORE scale. Conclusions: The ALTAIR tool shows a good reliability in the concordance inter observers, intra observer and inter device measurements and a concordant validity to show an association with vascular parameters, target organ damage and cardiovascular risk

    Status and Recent Results of the Acoustic Neutrino Detection Test System AMADEUS

    The AMADEUS system is an integral part of the ANTARES neutrino telescope in the Mediterranean Sea. The project aims at the investigation of techniques for acoustic neutrino detection in the deep sea. Installed at a depth of more than 2000m, the acoustic sensors of AMADEUS are based on piezo-ceramics elements for the broad-band recording of signals with frequencies ranging up to 125kHz. AMADEUS was completed in May 2008 and comprises six "acoustic clusters", each one holding six acoustic sensors that are arranged at distances of roughly 1m from each other. The clusters are installed with inter-spacings ranging from 15m to 340m. Acoustic data are continuously acquired and processed at a computer cluster where online filter algorithms are applied to select a high-purity sample of neutrino-like signals. 1.6 TB of data were recorded in 2008 and 3.2 TB in 2009. In order to assess the background of neutrino-like signals in the deep sea, the characteristics of ambient noise and transient signals have been investigated.     The ANTARES Telescope Neutrino Alert System

    The ANTARES telescope has the capability to detect neutrinos produced in astrophysical transient sources. Potential sources include gamma-ray bursts, core collapse supernovae, and flaring active galactic nuclei. To enhance the sensitivity of ANTARES to such sources, a new detection method based on coincident observations of neutrinos and optical signals has been developed. A fast online muon track reconstruction is used to trigger a network of small automatic optical telescopes.
